The DRSC 2.0 is a collection of dsRNAs for genome-wide RNAi knockdown. It improves upon version 1.0 in several ways, most significantly in terms of minimizing the potential for off-target effects due to sequence similarity to genes other than the target. The library is regularly updated to accommodate newly identified genes and updated gene models. Features include: Availability. The DRSC 2.0 is available for on-site screening. In addition, a limited number of amplicons can be requested as PCR-amplified DNA The DRSC Collection for Hit Validation (DRSC Validation) DRSC Validation is a collection of amplicons for RNAi knockdown of genes that have previously scored as hits in any screen at the DRSC. As of spring 2008, DRSC Validation covered about 6,000 amplicons. Amplicons directed against unrepresented genes are generated when new lists of screen hits submitted by screeners. Our ultimate goal is for genome-wide coverage. Importantly, the DRSC Validation amplicons are distinct from the amplicons in the 2.0 amplicons (i.e. target a different region of the gene), providing independent validation of a positive RNAi result. Please note that confirmation of the RNAi result does not eliminate the need for other follow-up, such as in vivo confirmation of biological relevance. Availability. Screen hits can be requested as "cherry picks" from the DRSC Validation collection. The requested amplicons are provided as PCR-amplified DNA templates. These can be used for dsRNA production at the home institution.
