Dr. Frank serves as the president of Pure Protein and Pure MHC, LLC. He has experience in small molecule and biologics drug development spanning research, discovery, preclinical, and clinical development. He has expertise in variety of therapeutic areas including immunology, inflammation, oncology, infectious disease, and autoimmune disease.

We provide the Non-human primate (NHP) research community with information on how these monkeys MHC function and present peptides with a view toward finding macaque MHCs that are homologues of human MHC proteins.

Dr. Buchli joined Pure Protein L.L.C. in 2001 and is currently applying his expertise in Immunology and Biochemistry as the Director of Research. His work is essential to the completion of ongoing research to develop new methodologies and technologies for developing novel soluble HLA proteins for one of the foremost biomedical companies in the country. Dr. Buchli’s past and present work has focused on HLA. The research of HLA is increasingly important to gaining a better understanding and finding improved treatments of infections and diseases including HIV and cancer.

Antibodies to HLA proteins are found in the blood supply. A blood recipient can suffer and die from transfusion related acute lung injury (TRALI) when antibodies to HLA in the blood they receive causes their lungs to fail. It is therefore likely that the blood supply will be screened for antibodies to HLA. Pure is working with the blood banking industry to develop an assay that will detect antibodies to HLA in donated blood.

HLA proteins act as an alarm system for cancerous and infected cells, with T-Cells being the first responders. When T-Cells recognize and bind to a foreign HLA epitope, they trigger the alarm and produce antibodies. This in turn amplifies the response to the diseased cells presenting the foreign HLA epitope. Similar to infections, responses to foreign HLA can be triggered when a person is exposed to these HLA alleles through transplants, child birth, or transfusion. This can cause the cascade effect that produces antibodies directed against foreign HLA, leading to rejection.
