Syndax Pharmaceuticals is a phase 2 stage oncology company developing combination therapies based on epigenetic modulation as a way to extend the benefit of proven cancer therapies. Targeted therapies represent an important class of agents used to treat tumors based on blocking the tumor’s addiction to growth promoting signals. Primary among these is endocrine therapy- including drugs known as aromatase inhibitors- designed to block the production of estrogen which promotes the growth of breast cancer cells. Syndax’s lead product candidate, entinostat, is a novel histone deacetylase inhibitor that modulates chromatin structure to reprogram the genes which influence the reversible and epigenetically-based development of resistance to targeted therapies. Entinostat is in phase 2 clinical trials in breast cancer, non-small cell lung cancer and Hodgkin’s lymphoma. In collaboration with the National Cancer Institute(NCI), Syndax is pioneering the use of entinostat in combination with another epigenetic agent- known as DNA methyltransferase inhibitors- to target resistance with a ‘dual’ epigenetic approach in other solid tumors. The NCI also is supporting a number of other combination studies with entinostat in solid as well as hematologic malignancies.
